Counting malassez

Shake middle

Put some cells into ependorf from the pot

~ 20 mL of cells into a pit + 20 mL blue

-> cell malassez

N_malassez = Number of cells inside the 25 # Concentration initiale Cell (CiC) = N_malassez * 8000 / 1.000.000 M/mL
Concentration to reach (CtR) = 0.5 M/mL

Cell Volume to extract (Ce) = CtR/CiC * Final volume Middle volume = Final volume - Ce

Counting HL60 P0 = 167 –> 1.336 M/mL
Counting MOLM14 P0 = 31 –> 0.248 M/mL

First subculture of HL60

Dilution from 1.336 M/mL to 0.5 M/mL in 10mL
–> Ce = 0.5/1.336 * 10 mL = 03.7 mL
–> MV = 10 - 03.7 = 6.3 mL

Counting HL60 P1 = 97 –> 0.776 M/mL Counting MOLM14 P0 = 65 –> 0.52 M/mL

Second subculture HL60

Dilution from 0.776 M/mL to 0.3 M/mL in 10mL
–> Ce = 0.776 / 0.3 * 10 mL = 26 mL
–> MV = 10 - 0.37 = 0.63 mL
